This document specifies a horizontal method for - the detection of L. monocytogenes, and - the detection of Listeria spp. (including L. monocytogenes).This document is applicable to - products intended for human consumption and for the feeding of animals, and - environmental samples in the area of food production and food handling. It is possible that certain additionally described Listeria species may not be detected or confirmed by this method. Within the scope of this document four consecutive steps are required for the detection of L. monocytogenes and Listeria spp.: 1. an initial enrichment in a selective liquid enrichment medium with reduced concentrations of selective agents (half-Fraser broth); 2. a second enrichment in a selective liquid enrichment medium with full concentrations of selective agents (Fraser broth); 3. plating on Listeria agar according to Ottaviani and Agosti and any other solid selective medium, as well as identification; 4. presumptive Listeria colonies are subcultured and confirmed using appropriate morphological and/or biochemical tests. Standards committee NA 057-01-06 AA "Mikrobiologie der Lebensmittelkette" ("Microbiology of the food chain") at DIN Standards Committee Food and Agricultural Products (NAL) is responsible for the standard.
